Losing hard drive space

I am losing hard drive space rapidly for no reason.
I tried disk cleaning, defragmenting and even removed a few programs and files but the problem persists.
Of the 144 GIGS on my C-Drive, I now only have approximately 2.5 GIGS available. Please help.
